History of AROMASIN:
Exemestane is a steroidal inhibitor used in postmenopause women who are adjuvant for treating breast cancer (also called hormonally sensitive, oestrogen responsive) in oral steroidal aromatase. It binds to the active site of the enzyme in an irreversible manner, leading to persistent inhibition.
Chemical Formula of AROMASIN:
Indication of AROMASIN:
To treat advanced breast cancer in postmenopausal women whose illness has progressed after tamoxifen therapy.
Mechanism of action of AROMASIN:
The development of breast cancer cells may be estrogen-dependent. Aromatase is the primary enzyme responsible for converting androgens to estrogens in both pre-and postmenopausal women. While the ovary is the primary source of oestrogen (mainly estradiol) in premenopausal women, adrenal conversion is the primary source of circulating estrogens in postmenopausal women. Deprival of oestrogens in conjunction with suppression of aromatase treatments are successful and selective in some women postmenopausal with hormonal breast cancer. Exemestane is a steroidal aromatase inactivator that is structurally similar to the natural substrate androstenedione. It binds to the active site permanently, producing permanent inhibition and required de novo synthesis to restore enzymatic function. Exemestane significantly lowers circulating oestrogen levels in postmenopausal women while having no effect on corticosteroid or aldosterone synthesis in the adrenal gland. This decrease in serum and tumour oestrogen concentrations slows tumour development and disease progression.
How to use AROMASIN:
The suggested dose of AROMASIN is one 25 mg tablet once daily after meal in early and advanced breast cancer.
- Postmenopausal women with estrogen-receptor positive early breast cancer who have had two to three years of tamoxifen and are switched to AROMASIN for a total of five years of adjuvant hormonal therapy.
Side effects of AROMASIN:
Aromasin may cause the following adverse effects: swelling in your hands or feet, chest pain, new or unusual bone pain, abrupt numbness or weakness, shortness of breath, and nausea.
